Simvastatin Attenuates Monocrotaline-induced Pulmonary Hypertension in Rats

Abstract

|
Pulmonary hypertension is characterized by abnormal proliferation of vascular endothelial and smooth muscle cells, leading to right heart failure and death. Simvastatin confer potent antiproliferative and antiinflammatory cardiovascular benefit. We hypothesized that simvastatin would attenuate pulmonary hypertension induced by monocrotaline (MCT) in rats. Twenty one 8-week-old rats were randomized to comtral, MCT (60 mg/kg, sc) and MCT plus simvastatin (5 mg/kg/day, po) treatment groups. Four weeks later, right ventricular (RV) pressure, right ventricular hypertrophy and pulmonary artery morphometry were measured. In the MCT group, RV pressure, right ventriculat hypertrophy and medial wall thickness of peribronchiolar artery and pulmonary arterioles were significantly increased (p<0.05), however, shich were markedly suppressed in the simvastatin treatment group (p<0.05). In this study, we speculate that chronic treatment of simvastatin may provide an additional strategy for the treatment of pulmonary arterial hypertension.
